Run the System File Checker tool (SFC.exe). To do this, follow these steps:
1. Open an elevated command prompt. To do this, click Start, click All Programs, click Accessories, right-click Command Prompt, and then click Run as administrator. GfxUI.EXE is not a valid win32 applicationOk, I just built my first system and everytime I turn on the computer I get GfxUi.exe is not a valid win32 application. I downloaded the new driver 6.14.10.5225 and it worked for a night and then it went back to the gfxui popping up on start up. It does not seem to effect anything, but is anyone else having this problem?
I have a core i3 530, gigabyte h55m-s2h mobo, winXp pro sp 2. I am overclocking it, but have turned it down till I find out what this is about. Thank you in adance.

I ran into this problem during a Windows 7 build this week. Turns out it IS a user permissions issue as is mentioned on many forums, although the reg hack that most folks specify did not work for me. Instead I took a look at the security tab for the file, and worked my way backwards.

I received this error after installing the Intel Corporation - display - Intel(R) HD Graphics driver, shown on the Microsoft Updates 'Recommended' list as Intel HD Graphics A03. For those trying to cross reference, I am using a Dell Latitude E5510 with Windows 7 Pro installed. This error occurred using both Dell BIOS A03 and A06.
One peculiarity that guided my search was that the error only occurred when I used my created local administrator account. When I used a standard user account, or the default Administrator account, no error.

Enable, then log in as the default Administrator account (to prevent possible security issues when using created local user account). Go to the %root%WindowsMicrosoft.NETFrameworkv2.0.50727 directory. RC the Config directory and select Properties. Select the Security tab, and click the Advanced button. Change ownership from 'Trusted Installer' to the default Administrator account. Go back to Security tab and click Edit. Change permissions for Administrators group to 'Read & Execute'.

Solved - I had the problem on a lenovo thinkpad with win7 64bit - German localization.
The fix with adjusting the permissions as described by marktinder worked for me as well. Maybe this is of help for users encountering the same 'GfxUi not working' problems on lenovo thinkpads.

If you get GFXUI.exe error on startup saying CLR error you can make it stop by RCing on the .exe file win system32 and setting it to compatibility mode for 95. Works for me, and I tried installing .net framework, reinstalling, flat out deleting the file, reinstalling my intel graphics driver, nothing else worked.
